Google Penguin 4.0 Update is not like at all like an old school, Now its going to be a real time, once your website will be crawl by google it will reflect the changes at the same time, if you have done anything spam (in terms of collecting backlink unethically) then you will hit by the update and suppose once you will disavow all the spamy link, then you will get the clean cheat once google will crawl your website again.

Also this time it will not going to affect the whole site, instead he will penalize the specific page for which spam has been done..
